N1-甲基假尿苷(N1-Methylpseudouridine)是一種修飾核苷,用于增強(qiáng) mRNA 翻譯。將N1-甲基假尿苷摻入 mRNA可關(guān)閉免疫/eIF2α 磷酸化依賴性翻譯抑制,并增加mRNA上的核糖體的密度,從而顯著促進(jìn)翻譯過程。N1-甲基假尿苷在翻譯能力方面優(yōu)于其他幾種修飾的核苷和相應(yīng)的組合。
輝瑞Pfizer-BioNTech和莫德納Moderna開發(fā)的COVID-19 mRNA疫苗證明了N1-甲基假尿苷修飾在mRNA疫苗開發(fā)中的出色成果,其中尿苷在兩種mRNA疫苗中完全被N1-甲基假尿苷取代。N1-甲基假尿苷修飾的RNA的制備是通過T7 RNA 聚合酶介導(dǎo)的體外轉(zhuǎn)錄酶促摻入相應(yīng)的N1-甲基假尿苷三磷酸(N1-Methylpseudo-UTP)來進(jìn)行的。
